The advert states that drop one will go live tomorrow, so you will have to wait a few hours until you see it.
The others will drop every 4 weeks or so (read the news page - "You'll be able to obtain each drop via a personalized, one-time-use promo code").

If you already have Prime, do you get the rewards anyway? I have prime, and while I don't really need the rewards I am just curious whether it's an additional subscription beyond just having Prime

Yes follow the link, you'll already have Prime gaming, it will give you a code with a copy button, paste it in your promo code box and get the reward
Great if you have Prime - if you don't I don't think the rewards are anywhere near enough to encourage players to sign up for Prime
