If a siege is already ongoing, and I remove the equipped gear from a champion that is set in a defensive slot:
1) the equip change will affect the champion in case it is attacked by the opponent team (example, I remove SPD boots, now that champion will be slower)
2) the equip change won't affect the champion which will keep the gear/stats the champ had at the time the siege matchmaking was done
Their stats are locked to whatever gear they were wearing at the moment siege prep ended and matchmaking began.

Probably in an announcement somewhere on Discord. I've just stuck by the set my gear/presets right before sleep and don't touch anything until after it starts for awhile now.
I did this last Siege and it worked like a charm. Didn't need to do it for the defenses I ran this Siege.
Now that free gear removal is permanent, no reason not to do it when needed.
IIRC, the "stats are locked once match-making begins" was in an announcement way back when Siege was introduced, but I don't think it is mentioned in the in-game info pop-up. That would be too logical.
