Yes, 6 star makes the most difference, although for power numbers of 45k to 60k, you'd also need endgame gear (legendary 6 star gear). However, don't look too much at the power ratings, they can be quite misleading. A champion or a team with high speed and the right skillset could easily defeat a champion/team with twice as much power. 

60’s Along with gear and my biggest mistake I made was ignoring masteries past the first 100 scrolls.  I was too involved with trying to complete missions and progress in dungeons.  Big mistake Finally got my tayrel with warmaster after 3 months and increased my clan boss damage by roughly 1.5 million. I can’t wait to see what my clan boss damage is once I get my team full masteries.   

Someone else mentioned  in another thread which is what I do now.  When I have XP boost I will only use my energy to complete daily quests and level champs/food.  When I don’t have XP boost I will do Daily’s, Dragon and Minotaur 

Don’t spend gems on shard packs.
